چکیده

Prior work has shown that mild alkaline hydrolysis of nuclei previously extracted with cold dilute acid and with lipid sol vents yielded not only the expected RNA nucleotides but also free adenine and guanine in amounts which varied in versely with the extent of preliminary lipid extraction. The nucleic acid of the nuclei was identified as the source of the free purines. The experiments described here demonstrated that the DNA of the nuclei was the source of the free adenine and probably also of the guanine, and that the conditions to which the nuclei had been subjected were such that significant amounts of purines could not have been removed from isolated DNA. The combined results support the hypothesis that the DNA contains purine deoxyribotides with direct or indirect linkages to lipid which render the deoxyribosidic bonds labile to hy drolysis following extraction with lipid solvents. The lymphosarcoma was found to have a high nuclear ribonucleic acid content when compared with thymus and myo cardium. The cytoplasmic and nuclear ribonucleic acids of the lymphosarcoma were unusual in that each had a high cytidylic acid content. In the analyses of the base content of the DNA, statistically significant differences between lymphosarcoma and thymus were found with respect to adenine and thymine.

A chromatographic comparison of the nucleic acids from isologous newborn, adult, and neoplastic thymus.

The total tissue nucleic acids (DNA and RNA) have been isolated from normal and neoplastic mouse thymus glands and chromatographically fractionated. Significant differences in the chromatographic distribution of the nucleic acids from normal adult thymus and x-radiation-induced thymic lymphosarcoma were found. Chromatographic patterns intermediate between normal and tumor were found in the thym...

Effect of Helium-Neon Laser and Sodium Hypochlorite on Calf Thymus Double-Stranded Deoxyribonucleic Acid Molecule: An in Vitro Experimental Study

Introduction: Low-energy helium-neon (He-Ne) laser beam lightis used in combination with sodium hypochlorite (Na2HOCl3) for clinical purposes. Regarding this, the present study aimed to investigate the effect of He-Ne laser (632.8 nm) and sodium hypochlorite on the calf thymus double-stranded deoxyribonucleic acid (ctdsDNA) molecule.  Materials and Methods: For the purpose of the study, ctdsDNA...

Distribution of 5-methylcytosine in the DNA of somatic and germline cells from bovine tissues.

Genomic DNA of calf thymus contains 1.5 times as much 5-methylcytosine as similar sperm DNA, but the major EcoRI repeat fragment from satellite I of thymus contains ten times as much 5-methylcytosine as the corresponding fragment from sperm DNA. Restriction enzyme analyses of the total DNA and the satellite I fragment show that three HpaII sites in the fragment are completely unmethylated in sp...

Identification of delta helicase as the bovine homolog of HUPF1: demonstration of an interaction with the third subunit of DNA polymerase delta.

Delta helicase is a 5' to 3' DNA helicase that partially co-purifies with DNA polymerase delta (pol delta) from fetal bovine thymus tissue. We describe the resolution of delta helicase from pol delta on heparin-agarose chromatography and its purification to apparent homogeneity by affinity purification on single-stranded DNA-cellulose chromatography, unique-sequence RNA-agarose chromatography, ...
